Been using my TF300T for a couple months, but all of a sudden last night my screen would not rotate. Anybody have any idea how to fix this?

You may have disabled it.
In the notifications bar there is a little icon, looks like a rectangle with arrows around it that enables or disables rotation.

And if that doesn't do it, you could try a cold boot (link in signature).
But you might also find that the only thing that gets it working 100% of the time it happens, if you haven't turned off rotation is to turn the tablet OFF & leave it off for 30-60 minutes. This happened a few times on my tf101 - no rhyme nor reason - & that was the only "trick" that reliably worked for me.

Go to Settings>Accesibility and enable Auto Rotate Screen.
