Cold boot doesn't work for me either. I'm going to try uninstalling apps one at a time to see if it is an app conflict

You know, plugging in headphones in fact is a work-around.

The issue does not appear to manifest in any form when the headphones are plugged in. Issue returns shortly after headphones are removed.

    Sound stops working on ICS

Have you found anything out about the sound loss?

I only recently purchased a TF101 (loving it) and once I'd bought the keyboard dock it has now started to do what you mentioned. Randomly the sound will stop and the only thing I can do to get it back is to reboot the device (normal shut down and power up).

I've been trying to work out what might have caused it but have come up blank. I don't have any volume control apps installed, and in fact only have a few apps on the device at the moment (Nova Launcher Prime, SugarSync, Dolphin Browser, Hackers Keyboard, Evernote, Kindle, Comics, Dark Horse, BBC IPlayer, etc). I did disable a number of the stock ASUS apps that I won't be using:

    Asus Email (using GMail)
    ASUS MyZine
    PC Sync Launcher
    Email
    Exchange Services
File Manager (using ES File Explorer)
    Movie Studio
    MyCloud (using SugarSync)
    MyNET
    News and Weather
    PressReader
    Talk
    SuperNote
    TegraZone Games
    WebStorage
    Zinio

But I can't see that disabling any of those would affect the sound.

I'm also suffering from this really annoying niggle. I've just raised it with Asus along with my other ICS problems (reboots, auto-rotate intermittently not working) in the vain hope they may be able to help and / or are at least acknowledging the problem exists. Will update if there is anything useful that comes out of it.

Cold Boot: Unplug the power cord and shut the unit off and Press and Hold the power button and the "Down" volume button together until after the normal start up screen appears and is joined by some small print at the top of the screen then let them both go. Just wait and it will do its thing.

Seriously? Thanks ASUS and Google. How about we tap our shoes together, too, and chant, "there's no place like home, there's no place..."

And what a surprise... another solution doesn't work. Why do people make excuses for ASUS and Google? Why do people write and say that these issues can be fixed by "knowledgeable people" in 30 minutes? They can't be solved by God in 30 minutes!!!!

I brought this sound issue up in another thread that got terminated. There is just no excuse for an update to screw up sound... period. ASUS and Google need to be held accountable, and they will, through reduced sales numbers. The Apple juggernaut just steamrolls over everyone with no viable alternatives to their closed architecture. Darn.
On the other hand, an iPad doesn't lose sound when the iOS updates.

From my experiments over the last few days the sound cutting out only seems to occur if I'm using the keyboard dock. If the unit remains separate the sound seems to be OK. That said, I usually shutdown the device of an evening, but for me at least, using the dock does seem to cause sound issues.

Also, has anyone else noticed that connecting headphones does NOT mute the device speakers?!? Seems to make headphone use a little pointless.

I also noticed that the US has had a new firmware update .21, I'm in the UK and it isn't available yet, not sure if this update will help.

Mine has sound issues, docked or not does not seem to make any difference. But I have never experienced that the built-in speakers didn't mute when connecting my headphones.

I believe it might just be notification sounds that are played on all speakers regardless of headphones being connected.

Any updates on this at all?

I don't seem to have any change if I have the dock connected or not. Only matters if I have headphones connected or not.

I have had an odd situation where my alarm (played on all speakers) started playing only on my headphones after a couple minutes. It makes me wonder if it starts playing sounds as if headphones are plugged in when they are in fact not plugged in.
