No it will not work.
Knowing I was going to order the new Prime when it was available, I ordered the existing dock when the price dropped, because I had previously read in more than one place that "ASUS assures users that the original dock would work with the Prime." Now I see there is going to be a dock for the Prime. Does anyone know whether the original dock will or will not work with the Prime? It hasn't reached me yet and I can easily send it back to Amazon, but I wish I knew yes or no.

Wondering where you got the original information. It would appear there is enough of a size difference in the tablet itself that it probably wouldn't fit the original dock

I also read that they were keeping it close enough that the original dock would be compatible with the transformer2. I'm kinda PO'd that they did not hold true to their announced plan. It was in an official website, but I can't remember what one. Maybe they figured with so many problems the dock had to start with, they were starting from scratch.

With the way rumors have flown around there are few things certain. Detection did (after a ton of searching) come up with actual specs that cleared some things up, like the fact that the sound is stereo but there's only one speaker. Go figure.

I remember reading it in the flurry of articles that appeared after the press conference a bit ago (pre-Nov 9th), that "Asus officials" had verified that the original dock would work with both products. I read it more than once. Guess they wanted another way to make money ;-( I'm actually thinking I may not get one now. I believe the specs are changed a little too. I won't be desperate to get one, at any rate.
I did call Asus tech support (India...) and they told me the old dock wouldn't work.
